General description

The 5′→3′ polymerase activity of Taq DNA Polymerase and the 3′→ 5′ exonuclease of the proofreading polymerase combine for improved performance and results; 3′→5′ exonuclease proofreading produces a threefold increase in DNA synthesis fidelity compared to Taq DNA polymerase.
PCR products have both blunt ends and 3′ single A-overhangs just like using Taq DNA polymerase, ideal for blunt end cloning or T/A cloning.
The High Fidelity PCR Master is a ready-to-use, double-concentrated master mix that contains all the reagents required to perform PCR (except template and primer). Water, PCR Grade, is also supplied.
This powerful specific enzyme blend of the High Fidelity PCR Master is designed to give PCR products with high yield, high fidelity, and high specificity from episomal and genomic DNA.
The special enzyme blend is composed of two thermostable polymerases: Taq DNA Polymerase and a thermostable polymerase with proofreading activity. Due to the inherent 3′-5′ exonuclease activity of the proofreading polymerase, the High Fidelity PCR Master results in approximately three times the fidelity and approximately twice the yield of Taq DNA Polymerase alone.

Choose the convenient High Fidelity PCR Master, a double-concentrated, ready-to-use mix, which combines Expand High Fidelity PCR System with PCR-Grade dNTPs, MgCl2, and an optimized reaction buffer. The High Fidelity PCR Master delivers twice the yield and three times better fidelity than Taq DNA Polymerase alone.High yield and increased fidelity compared to Taq DNA Polymerase. Use the High Fidelity PCR Master for:

  • PCR up to 5 kb
  • RT-PCR up to 5 kb

High Fidelity PCR Master is stable at +2 to +8°C, allowing PCR without time-consuming reagent thawing.

Each lot is PCR function tested using human genomic DNA and primers specific for a 4.8 kb fragment of the tPA gene.
