應用

内-β-半乳糖苷酶已用于分级蛋白质分离。它可用于人内皮细胞内皮分泌组的糖蛋白质组中的去糖基化。

生化/生理作用

未分枝的重复聚乙酰乳糖胺中的内部β(1-4)半乳糖连接[GlcNAc β(1-3)Gal β (1-4)] 结构是优选的底物。

诸如硫酸角质素的硫酸化结构也会被裂解。底物的支化或岩藻糖基化可减少或完全抑制裂解。半乳糖上C-6的硫酸化将阻断裂解。取决于与优选底物的偏差,新乳酸基团的低聚糖会以极低的速率而被裂解。例如,Gal β(1-3)GlcNAc β(1-3) Gal β(1-4)Glc会以硫酸角质素5X10-5的速率而被裂解

β-半乳糖苷酶可将乳糖切割成单糖成分、葡萄糖和半乳糖。它还在反馈环中催化葡萄糖转糖基化为β-半乳糖苷酶的诱导物异乳糖。

單位定義

在pH 5.8、37 °C条件下,一单位酶每分钟可从牛角质角蛋白硫酸盐中释放1.0 μ摩尔的还原性糖。
