5,5′双二硫代(2-硝基苯甲酸)也称为DTNB或Ellman′s试剂。这是一种与巯基相互作用的水溶性试剂,在中性pH下方便地应用。硫代阴离子(RS-)与Ellman′s试剂(DTNB2-)反应生成2-硝基-5-硫代苯甲酸盐阴离子(TNB2-)和一种混合二硫化物(R-S-TNB-)。TNB2- 具有较高的摩尔吸收系数,在412nm处有吸收。这种吸收与pH无关,在pH>7.3。

應用

Ellman′s试剂可用于乙酰胆碱酯酶的检测,并可对半胱氨酸残基进行特异性标记。该试剂是测定蛋白质、多肽和组织中游离巯基含量的敏感试剂。
