成分

酵母转化试剂盒包含:
  • 转化缓冲液;100 ml;100 mM 醋酸锂、10 mM Tris HCl (pH 7.6) 和 1 mM EDTA
  • 平板缓冲液;100 mL;40%PEG,100 mM 醋酸锂,10 mM Tris HCl,pH 7.5,1 mM EDTA
  • 鲑鱼试验的脱氧核糖核酸,10 mg/mL;2 x 1 mL
  • 携带 ura 基因的对照酵母质粒 DNA pRS316;10 μg
  • 不含尿嘧啶的酵母合成脱落培养基补充剂;1 g

原則

用补充突变基因的质粒转化使转化子在缺乏所需成分的培养基上生长。通过在缓冲醋酸锂溶液中孵育,使酵母细胞具有转化能力。然后在含聚乙二醇 (PEG) 的溶液中将细胞与转化 DNA 和载体 DNA 一起孵育进行转化。

Transformation is the process by which exogenous DNA is introduced into a cell, resulting in a heritable change or genetic modification. This was first reported in Streptococcus pneumoniae by Griffith in 1928. Transforming principle of DNA was demonstrated by Avery et al. in 1944.
