磷酸果糖激酶 (PFK) 催化糖酵解的第三步,即果糖-6-磷酸转化为果糖-1,6-二磷酸,即糖酵解的限速步骤。PFK 活性除转化后修饰外,还受多种辅助因子的高度调控,可用于测量组织中的糖酵解通量。缺乏 PFK 活性可导致糖原贮积病,糖原贮积病类型 II 型(Tarui 病)——一种糖原贮积病。

原則

磷酸果糖激酶比色测定试剂盒为测定多种样品中的 PFK 活性提供了一种简单直接的程序。通过偶联酶试验测定 PFK 活性,其中果糖-6-磷酸和 ATP 通过 PFK 转化为果糖-1,6-二磷酸和 ADP。ADP 被酶混合物转化为 AMP 和 NADH。生成的 NADH 还原无色探针,生成与 PFK 活性成比例的比色 (450 nm) 产物。PFK 的 1 个单位是 37°C、pH 7.4 时每分钟生成 1.0 μmol NADH 的酶量。
