亮肽素或N-乙酰基-L-亮氨酰-L-亮氨酰-L-精氨酸是天然存在的三肽,尤其适合用作丝氨酸蛋白酶抑制剂和半胱氨酸蛋白酶抑制剂。 其作用机理涉及结构相似的共价结合反应:
- 在丝氨酸蛋白酶的活性位点中,亮肽素在亮肽素的醛基和酶活性位点丝氨酸残基的羟基之间形成共价半缩醛加合物。
- 在半胱氨酸蛋白酶的活性位点中,亮肽素的亲电子(醛)碳与酶活性位点中半胱氨酸残基的硫原子形成相当的键。
生化/生理作用
丝氨酸和半胱氨酸蛋白酶抑制剂。抑制纤溶酶、胰蛋白酶、木瓜蛋白酶、钙蛋白酶和组织蛋白酶B。不抑制胃蛋白酶、组织蛋白酶A和D、凝血酶或α-糜蛋白酶。有效浓度为10-100 μM.
