一般說明
组蛋白是含有H3、H4、H2A和H2B的八聚体蛋白。它是DNA是围绕其缠绕形成核小体结构的核心蛋白。核心组蛋白包含球状结构域。每个蛋白在其N端均由碱性氨基酸组成。H2A在其C末端具有一个额外的氨基酸,形成组蛋白尾巴。核小体结构在染色质浓缩过程中通过接头组蛋白稳定。
應用
小牛胸腺中的组蛋白已用于:
- 在甲基转移酶反应混合物中进行甲基转移酶测定
- 在体外结合试验中,探讨了MORF4(4号染色体上的死亡率因子)-15号染色体上相关基因(MRG15)与组蛋白的潜在结合
- 在体外共培养试验中刺激肝脏驻留的Kupffer细胞
生化/生理作用
核心组蛋白参与组蛋白-组蛋白各种相互作用并维持核小体DNA的组织。组蛋白尾巴有助于形成更高阶的核小体结构。染色质纤维的整体结构和作用受组蛋白修饰的调节。
