Specificity

Clone 1F16 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Solute carrier family 12 member 4 (SLC12A4). It targets an epitope within 20 amino acids from the cytoplasmic domain with in the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 20 amino acids from the cytoplasmic domain within the C-terminal region of human Solute carrier family 12 member 4 (SLC12A4).

Application

Quality Control Testing

Evaluated by Immunohistochemistry (Paraffin) in Human pancreas tissue sections.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution of this antibody detected SLC12A4 in Human pancreas tissue sections.

Tested Applications

Affinity Binding Assay: A representative lot of this antibody bound the recombinant fragment of Human SLC12A4 with a KD of 1.4 x 10-7 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected SLC12A4 in HeLa and K562 cells.

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected the recombinant fragment of Human SLC12A4.

Enzyme Immunoassay ELISA) Analysis: A serial of dilutions from a representative lot detected recombinant fragment of Human SLC12A4.

Target description

Solute carrier family 12 member 4 (UniProt: Q9UP95; also known as Electroneutral potassium-chloride cotransporter 1, Erythroid K-Cl cotransporter 1, hKCC1) is encoded by the SLC12A4 (also known as KCC1) gene (Gene ID: 6560) in human. Potassium-coupled chloride transporters (KCCs; SLC12A) play crucial role in regulating cell volume and intracellular chloride concentration. SLC12A4 is a homomultimeric, multi-pass membrane protein with seven cytoplasmic domains and twelve transmembrane domains. It can also form heteromultimers with other K-Cl cotransporters. It is ubiquitously distributed with higher expression observed in erythrocytes from patients with hemoglobin (Hb) SC and Hb SS compared to normal AA erythrocytes. This contributes to red blood cell dehydration and to the manifestation of sickle cell disease by increasing the intracellular concentration of Hbs. SLC12A4 mediate electroneutral potassium-chloride co-transport when activated by cell swelling. It is involved in volume homeostasis in single cells. Seven different isoforms of SLC12A4 have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
