Specificity

Clone 1F17 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Histone H2B ubiquitylated on lysine 120.

Immunogen

KLH-conjugated linear peptide corresponding to 13 amino acids surrounding ubiquitylated lysine 120 of human Histone 2B.

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell acid extract.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Ubiquityl Histone H2B-Lys120 in HeLa cell acid extract.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Ubiquityl Histone H2B-Lys120 in HL-60 cell and Rat2 cell acid extracts .

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Ubiquityl Histone H2B-Lys120 in human liver tissue sections.

Peptide Inhibition Assay: Target band detection in a lysate from HeLa cell acid extract was prevented by pre-blocking of a representative lot with Ubiquityl Histone H2B-Lys120 peptide, but not the corresponding non- Ubiquityl H2B peptide.

Affinity Binding Assay: A representative lot of this antibody bound Ubiquityl Histone H2B-Lys120 peptide with a KD of 3.5 x 10-8 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Histone H2B type 1-B (UniProt: P33778; also known as H2B-clustered histone 3, Histone H2B.1, Histone H2B.f (H2B/f) is encoded by the H2BC3 (also known as H2BFF, HIST1H2BB) gene (Gene ID: 3018) in human. Histone H2B is a core component of nucleosome that wrap and compact DNA into chromatin, limiting DNA accessibility to the cellular machineries which require DNA as a template. Histones play a central role in transcription regulation, DNA repair, DNA replication and chromosomal stability. DNA accessibility is regulated via a complex set of post-translational modifications of histones, also called histone code, and nucleosome remodeling. Histone H2B features a main globular domain and a long N terminal tail. Histone H2B mono-ubiquitinated on lysine120 during transcriptional activation by RAD6E2 protein and is associated with the transcribed regions of highly expressed genes. It is a key histone modification that plays critical roles in gene transcriptional regulation and higher order chromatin organization. Mono-ubiquitination of histone H2B stimulates transcriptional elongation by facilitating FACT-dependent chromatin remodeling. De-ubiquitination of histone H2B lysine120 by USP22 is a required step in transcriptional activation. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
