Specificity

Clone 1C3 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects ARID domain-containing protein 1A (ARID1A). It targets an epitope within 22 amino acids from the internal region. May detect all three isoforms.

Immunogen

KLH-conjugated linear peptide corresponding to 22 amino acids from the internal region of human ARID domain-containing protein 1A (ARID1A).

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate..

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected ARID1A in HeLa cell lysate.

Tested applications

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected ARID1A in HeLa, A431, HUVEC and NIH 3T3 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected ARID1A in human colon tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound ARID1A with a KD of 4.3 x 10-8 in an affinity binding assay.

Target description

AT-rich interactive domain-containing protein 1A (UniProt: O14497; also known as ARID domain-containing protein 1A, B120, BRG1-associated factor 250, BAF250, BRG1-associated factor 250a, BAF250A, Osa homolog 1, hOSA1, SWI-like protein, SWI/SNF complex protein p270, SWI/SNF-related matrix-associated actin-dependent regulator of chromatin subfamily F member 1, hELD) is encoded by the ARID1A (also known as BAF250, BAF250A, C1orf4, OSA1, SMARCF1) gene (Gene ID: 8289) in human. ARID1A is a member of a large family of proteins that contain a characteristic ARID domain of approximately 100 amino acids that binds DNA fragments rich in adenine-thymine (AT). It is a component of SWI/SNF chromatin complexes and is highly expressed in spleen, thymus, prostate, testis, ovary, small intestine, and colon. At lower level it is also expressed in several other tissues. It is involved in transcriptional activation and repression of select genes by chromatin remodeling. It changes chromatin structure by altering DNA-histone contacts within a nucleosome in an ATP-dependent manner. ARID1A is shown to be critical for embryonic stem cell pluripotency and their differentiation into mesoderm-derived cardiomyocytes and adipocytes. It is also a component of the neural progenitors-specific chromatin remodeling complex (npBAF complex) and the neuron-specific chromatin remodeling complex (nBAF complex). During neural development a switch from a stem/progenitor to a postmitotic chromatin remodeling mechanism occurs as neurons exit the cell cycle and become committed to their adult state. Nuclear ARID1A is shown to be more unstable than its cytoplasmic counterpart, as it degrades rapidly, by the ubiquitin-proteasome system of the nucleus. It is normally detected during the G0-G1 phase of the cell cycle and its levels decrease during the G2-S phase. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
