Specificity

Clone 2D17 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Contactin-associated protein-like 2 (CNTNAP2). It targets an epitope within 17 amino acids from the C-terminal, cytoplasmic domain.

Immunogen

KLH-conjugated linear peptide corresponding to 17 amino acids from the C-terminal region of human Contactin-associated protein-like 2 (CNTNAP2).

Application

Quality Control Testing

Evaluated by Western Blotting in Rat brain tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected CNTNAP2 in Rat brain tissue lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected CNTNAP2 in Human brain tissue lysate.

Affinity Binding Assay:: A representative lot of this antibody bound CNTNAP2 peptide with a KD of 3.7 x 10-8 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected CNTNAP2 in human cerebral cortex tissue sections.

Anti-CNTNAP2, clone 2D17 ZooMAb®, Cat. No. ZRB1079, is a rabbit recombinant monoclonal antibody that specifically targets CNTNAP2 and is tested for use in Affinity Binding Assay, Immunohistochemistry, and Western Blotting.

Target description

Contactin-associated protein-like 2 (UniProt: Q9UHC6; also known as Cell recognition molecule Caspr2, CNTNAP2) is encoded by the CNTNAP2 (also known as CASPR2, KIAA0868) gene (Gene ID: 26047) in human. CNTNAP2 is a single-pass type I membrane glycoprotein protein of the neurexin family that is predominantly expressed in the nervous system. It mediates synaptic cell adhesion and is required, with CNTNAP1, for radial and longitudinal organization of myelinated axons. It also plays a role in the formation of functional distinct domains that are critical for saltatory conduction of nerve impulses in myelinated nerve fibers. CNTNAP2 is synthesized with a signal peptide (aa 1-27), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 28-1262), a transmembrane domain (aa 1263-1283), and a cytoplasmic domain (aa 1284-1331). Knockdown of CNTNAP2 in cortical cultures is shown to impair the development of spines. Mutations in CNTNAP2 gene are known to be associated with intellectual disability, epilepsy, schizophrenia, language disorders, and autism. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
