Specificity

Clone 1D19 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Phosphohistidine phosphatase 1 (PHTP1).

Immunogen

His-tagged full-length recombinant human Phosphohistidine phosphatase 1 (PHPT1).

Application

Quality Control Testing

Evaluated by Western Blotting in HepG2 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected PHPT1 in HepG2 cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected PHPT1 in A549 cell lysate.

ELISA Analysis: 300 μg/mL from a representative lot detected PHPT1 protein in an indirect ELISA.

Affinity Binding Assay:: A representative lot of this antibody bound PHPT1 protein with a KD of 1.0 x 10-12 in an affinity binding assay.

Anti-PHPT1, clone 1D19 ZooMAb®, Cat. No. ZRB1056, is recombinant Rabbit monoclonal antibody that detects phosphohistidine phosphatase 1 (PHPT1) and is tested for use in Affinity Binding Assay, ELISA, and Western Blotting.

Target description

14 kDa phosphohistidine phosphatase (UniProt: Q9NRX4; also known as EC:3.9.1.3, Phosphohistidine phosphatase 1, PHPT1, Protein histidine phosphatase, PHP, Protein janus-A homolog) is encoded by the PHPT1 (also known as PHP14, CGI-202, HSPC141) gene (Gene ID: 29085) in human. Protein histidine phosphorylation is reported to exist widely in vertebrates and PHPT is the only phosphohistidine phosphatase described in mammals. It regulates phosphohistidine levels of several proteins including those involved in signaling, lipid metabolism, and potassium ion transport. It is expressed abundantly in heart and skeletal muscle and is also found in several other tissues. Human PHPT1 is composed of six β-strands that are flanked by two -helices. The substrate-binding site is shown to be positioned between two loops, one between 1 and β4, and the other between β5 and 2. This site contains multiple residues that are shown to be important for substrate binding and include Glu51, Tyr52, His53, Tyr93, and Met95. The active site His53, located at the beginning of helix 1, is identified as the catalytic residue and the NH groups of His53, Ala54, and Ala86 and the OH group of Ser94 act as anchors of substrate and stabilize phosphate binding to the active site. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
