Specificity

Reacts with the laminin receptor alpha-6beta-1 and alpha-6beta-4 complexes.

Application

Immunoprecipitation, flow cytometry, immunohistochemistry in frozen tissue sections. Product does not detect alpha 6 in Western blot since the epitope most likely involves both alpha 6 and beta 1 integrin subunit. Product does not block alpha 6 integrin mediated cell adhesion to laminin. Activation of integrin signaling properties by MA6 has not been characterized. Immunoprecipitation of VLA-6 from lysate of 10E6 cell equivalent requires 2 μg of MA6. Recommended concentration for flow cytometry - 10 μg/mL, staining of frozen tissue sections - 20 μg/mL.

Optimal working dilutions must be determined by end user.

Cell lines tested by using MA6 to express (6 integrins include: mouse bone marrow-derived mast cells, melanoma cell line B16F1, squamous carcinoma SCC-VII, fibroblast cell line NIH-3T3, splenic T lymphocytes, thymocytes and various T cell lines.
This Anti-Integrin α6 Antibody, clone MA6 is validated for use in FC, IP, IH for the detection of Integrin α6.

Schwann cells form basal laminae (BLs) containing laminin-2 (Ln-2; heterotrimer alpha2beta1gamma1) and Ln-8 (alpha4beta1gamma1). Loss of Ln-2 in humans and mice carrying alpha2-chain mutations prevents developing Schwann cells from fully defasciculating axons, resulting in partial amyelination. Integrin receptors mediate cell-extracellular matrix interactions and regulate many events, including cell growth, proliferation, and differentiation. Retinal integrins are incompletely understood, although these receptors are potentially important factors in normal retinal function and pathology. Immunocytochemistry was used to localize alpha
