Specificity

CBL419 is specific for eosinophil major basic protein irrespective of the stages of eosinophil activation and can be regarded as a "pan-eosinophil" marker. This antibody will recognize the 23.8 kDa secreated pro-form of MBP (Pregnancy Associated MBP) found in the placenta and serum of pregnant women as well as the 13.8 kDa mature form of the protein which is found in the matrix of the eosinophil large specific granual. It does not cross-react with horse, mouse or rat MBP.

Application

Anti-Eosinophil Major Basic Protein Antibody, clone BMK13 is an antibody against Eosinophil Major Basic Protein for use in ELISA, WB, IH(P).
Immunohistochemistry: Clone BMK-13 reacts with fresh frozen tissues that were sectioned (6μm), dried for 1 hour at room temperature and fixed in 1:1 solution of methanol and acetone, and airdried for 1 hour. Sections can be stored wrapped in aluminium foil at -70°C until used. For paraffin embedded sections, 4% PFA (3hr) should be used for skin biopsies. 5 μM sections are dewaxed, and rehydrated with water. A proteinase treatment of 0.1% trypsin for 15 minutes in water should be performed prior to blocking and antibody staining.The antibody reacts better in frozen tissues than in paraffin embedded tissues.
Western blotting: purified normal human eosinophils were treated with digitonin (5mg/mL) for 10 minutes before boiling in sample buffer and run on a 15% SDS-PAGE. Purified MBP migrates at ~ 14kDa, and typical human samples range from 10-16kDa.
ELISA development for MBP estimation.

Optimal working dilutions must be determined by the end user.

The monoclonal is presented at a concentration of 100μg/ml in phosphate buffered saline containing 10mM sodium azide and 1mg/ml bovine serum albumin. We recommend that each laboratory determine an optimum working titre for use in its particular application.

Storage and Stability

For use within 1 month of purchase store at +4°C, for long term storage aliquot antibody into small volumes and store at -20°C.
