General description

Serine/threonine-protein phosphatase 2A 56 kDa regulatory subunit gamma isoform (UniProt Q13362; also known as PP2A B subunit isoform B′-gamma, PP2A B subunit isoform B56-gamma, PP2A B subunit isoform PR61-gamma, PP2A B subunit isoform R5-gamma, Renal carcinoma antigen NY-REN-29) is encoded by the PPP2R5C (also known as KIAA0044) gene (Gene ID 5527) in human. Protein phosphatase 2 (PP2 or PP2A) is a ubiquitously expressed heterotrimeric serine/threonine protein phosphatase with broad substrate specificity, such as Raf, MEK, and AKT. PP2A is composed of a structural A and a catalytic C subunit as the dimeric core enzyme, and a regulatory B subunit. There exist two different A subunits (PPP2R1A & PPP2R1B), two different C subunits (PPP2CA & PPP2CB), and 12 different B subunits (PPP2R2B-D, PPP2R3A-C, PPP2R4, PPP2R5A-E). The enzymatic activity of a catalytic subunit is altered upon binding an A subunit even in the absence of a B subunit. The B subunits are believed to play key roles in controlling the localization and specific activity of the A/C holoenzymes.

Specificity

This antiserum is also known as M880 (Okamoto, K., et al. (1996). Mol. Cell. Biol. 16(11):6593-6602). Immunogen sequence is 100% present in human PPP2R5C spliced isoform 5 and Gamma-3, with the first amino acid missing in isoform 4 and Gamma-2. PPP2R5C spliced isoform Gamma-3 (UniProt Q13362-1) and Gamma-2 (UniProt Q13362-3) were originally referred to as B′α1 and B′α2, respectively (Tehrani, M.A., et al. (1996). J. Biol. Chem. 271(9):5164-5170). The Immunogen sequence is not present in PPP2R5C isoform Gamma-1 or any of the spliced isoforms of the other 11 types of human PP2A B subunits (PPP2R2B, PPP2R2C, PPP2R2D, PPP2R3A, PPP2R3B, PPP2R3C, PPP2R4, PPP2R5A, PPP2R5B, PPP2R5D, PPP2R5E).

Western Blotting Analysis: A representative lot detected the expression of exogenously transfected human PP2A B56γ subunit in HeLa cells, but not in cells co-transfected with B56γ shRNA (Lee et al., 2010; Zimmerman et al, 2012).
Western Blotting Analysis: A representative lot detected exogenously expressed human B′α1 (PPP2R5C spliced isoform Gamma-3) protein in SF9 insect cells and COS-1 cells, as well as the endogenous rat B′α1 following its isolation by immunopreciptation from a rat fibroblast cell line, clone 6 (Okamoto, K., et al. (1996). Mol. Cell. Biol. 16(11):6593-6602).
Immunocytochemistry Analysis: A representative lot detected differential PP2A B56γ3 subunit cytoplamic and nuclear distribution patterns by fluorescent immunocytochemistry staining of 4% paraformaldehyde-fixed, 0.1% Triton X-100-permeabilized HeLa cells at different stages of cell cycle (Lee, T.Y., et al. (2010). J. Biol. Chem. 285:21567-21580).
Immunoprecipitation Analysis: A representative lot immunoprecipitated exogenously expressed human B′α1 (PPP2R5C spliced isoform Gamma-3) from SF9 insect cells and COS-1 cells, as well as the endogenous rat B′α1 from a rat fibroblast cell line, clone 6 (Okamoto, K., et al. (1996). Mol. Cell. Biol. 16(11):6593-6602).

Quality

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:2,000 dilution of this antibody detected PP2A B subunit isoform B56-gamma in 10 µg of HeLa cell lysate.

Target description

~54 kDa observed. 56.66 kDa (isoform Gamma-2), 61.06 kDa (isoform Gamma-3), 62.76 kDa (isoform 4), 64.29 kDa (isoform 5) calculated. Uncharacterized band(s) may appear in some lysates.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
