General description

Tyrosine-protein kinase JAK1 (EC 2.7.10.2; UniProt P23458; also known as Janus kinase 1, JAK-1) is encoded by the JAK1 (also known as JAK1A, JAK1B) gene (Gene ID 3716) in human. JAK1 belongs to the family of non-receptor Janus tyrosine kinases (JAKs) that regulate a spectrum of cellular functions downstream of activated cytokine receptors in the lympho-hematopoietic system. Immunological stimuli, such as interferons and cytokines, cause the recruitment of Stat transcription factors to cytokine receptor. Subsequent phosphorylation by receptor-associated JAK causes Stat dimerization, nuclear translocation, and Stat-mediated target genes transcription. The canonical JAK/Stat pathway is integral to maintaining a normal immune system by stimulating proliferation, differentiation, survival, and host resistance to pathogens. Altering JAK/Stat signaling to reduce immune cells cytokine responses represents an attractive target for anti-inflammatory therapies. Although JAK1 is a ubiquitously expressed Janus kinase activated by a wide variety of cytokines, studies conducted on Jak1-knockout mice and cells indicate that JAK1 plays a non-redundant role in promoting biologic responses induced selectively by a subset of cytokine receptors.

Specificity

Predicted to react with a broad-range of mammalian species based on 100% sequence homology.
The Tyr1022/1023 designation is according to the JAK1 sequence originally published by Wilks, A.F., et al. (1991). Mol. Cell. Biol. 11(4):2057-2065 (PMID 1848670; NCBI accession: AAA36527). Numbering of the target pTyr residues varies depending on the species and source of the sequence. For example, target pTyr residues this rabbit polyclonal detects correspond to pTyr1034/pTyr1035 and pTyr1033/pTyr1034, respectively, of the human and mouse JAK1 sequence reported by UniProt (P23458 and P52332, respectively), or pTyr1033/pTyr1034 of bovine, canine, and rat JAK1 (NP_001193463 and NP_445918.1), pTyr1021/pTyr1022 of porcine JAK1 (NP_999279).

Dot Blot Analysis: A 1:1,000 dilution of this antibody detected 0.156-20 ng of the immunogen phosphopeptide, but not the corresponding non-phosphorylated peptide.
