Zastosowanie

Quality Control Testing

Evaluated by Western Blotting in K562 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Histamine Receptor 4 in K562 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected GST-tagged recombinant fragment corresponding to the third cytoplasmic domain of Human Histamine Receptor 4.

Immunohistochemistry (Paraffin) Analysis: A 1:100-1,000 dilution from a representative lot detected Histamine Receptor 4 in Human tonsil and Human dorsal root ganglion tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Histamine Receptor 4 in K562 cells.

Affinity Binding Assay: A representative lot of this antibody bound Histamine Receptor 4 peptide with a KD of 1.6 x 10-8 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Histamine H4 receptor (UniProt: Q9H3N8; also known as H4R, HH4R, AXOR35, G-protein coupled receptor 105, GPRv53, Pfi-013, SP9144) is encoded by the HRH4 (also known as GPCR105) gene (Gene ID: 59340) in human. Histamine, an endogenous short-acting biogenic amine, is synthesized from histidine through the action of histidine decarboxylase. Its actions are mediated via four subtypes of G-protein-coupled receptors, designated as H1, H2, H3 and H4 histamine. Histamine H4 receptor (H4R) is a multi-pass membrane glycoprotein with four extracellular domains, seven transmembrane domains, and four cytoplasmic domains. H4R mediates histamine signals in peripheral tissues and displays a significant level of constitutive activity. H4R is expressed on cells of the immune system and is also detected in a variety of peripheral tissues. Its expression is either up-regulated or down-regulated upon activation of the lymphoid tissues and this regulation may depend on the presence of IL-10 or IL-13. H4R m

Postać fizyczna

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Rekonstytucja

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
