Clone 1D7 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Chromobox protein homolog 1 (CBX1). It targets an epitope within 15 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 15 amino acids from the C-terminal region of human Chromobox protein homolog 1 (CBX1).

Zastosowanie

Quality Control Testing

Evaluated by Western Blotting in Caco-2 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected CBX1 in Caco-2 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected CBX1 in HeLa, L6, and NIH3T3 cell lysates.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected CBX1 in A431, HepG2, and NIH3T3 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected CBX1 in human testis tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound CBX1 peptide with a KD of 3.1 x 10-7 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Chromobox protein homolog 1 (UniProt: P83916; also known as HP1Hsbeta, Heterochromatin protein 1 homolog beta, HP1 beta, Heterochromatin protein p25, M31, Modifier 1 protein, p25beta, CBX1) is encoded by the CBX1 (also known as CBX) gene (Gene ID: 10951) in human. CBX1 is a homodimeric nuclear protein that is a component of heterochromatin. It is expressed in all adult and embryonic tissues. It recognizes and binds histone H3 tails methylated at Lys-9 via the chromo domain, leading to epigenetic repression. Also interacts with lamin B receptor (LBR) to contribute to the association of the heterochromatin with the inner nuclear membrane. CBX1 preferentially recognizes H3K9me3 (peptide binding specificity Kd = 5 M). However, substitution of threonine 6 to alanine reduces its affinity significantly (Kd = 45 M). CBX1 is reported to possess oncogenic activities in hepatocellular carcinoma (HCC). Its expression is significantly increased in HCC and higher expression has been linked to larger tumor size, poor tumor differentiation, and tumor vascular invasion. It interacts with transcription factor HMGA2 to activate the Wnt/β-Catenin signaling pathway. Suppression of β-Catenin is shown to abolish CBX1-mediated cell growth. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Postać fizyczna

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Rekonstytucja

30 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Przechowywanie i stabilność

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
