Cyclin-dependent kinase-like 5 (EC 2.7.11.22; UniProt O76039; also known as Serine/threonine-protein kinase 9) is encoded by the CDKL5 (also known as EIEE2, STK9) gene (Gene ID: 6792) in human. CDKL5, a proline-directed serine/threonine kinase that plays a role in regulating axon outgrowth, dendritic morphogenesis, and synapse formation in early postnatal life, and in maintaining synaptic function in the adult brain. It is detected in the nucleus and dendrites of neurons. Its expression appears to be low in the embryonic cortex and is strongly induced at perinatal and postnatal stages in maturing neurons. It is reported to shuttle between the cytoplasm and nucleus through a CRM1-mediated nuclear export mechanism. Its protein kinase domain is localized in amino acids 13-297 and the serine-threonine protein kinase active site resides in amino acids 127-144. It has large C-terminal extension of about 700 amino acids that has several functions. This region serves as a negative regulator of the catalytic activity and is also essential for its nuclear localization. Mutations in CDKL5 gene have been linked to developmental and epileptic encephalopathy 2 (DEE2), a severe form of epilepsy characterized by seizures or spasms beginning in infancy. Two isoforms of CDKL5 have been described that are produced by alternative splicing. This ZooMAb^® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material.
