Application

TAMRA-a-TCO is an axial trans-cyclooctene (a-TCO) for bioconjugation and protein labeling. As demonstrated by the Joe Fox lab in Pigga, et al, a-TCOs are more hydrophilic and reactive than standard TCOs. The fluorescent TAMRA-a-TCO was developed for rapid, live-cell and in vivo experiments. It is cell permeable, reacts with tetrazines intracellularly via Diels-Alder chemistry, and exhibits reduced background fluorescence relative to other TAMRA TCO tags when imaged using fluorescence microscopy (illumination at λ=531 nm).

Other new a-TCOs:
  • NHS ester-a-TCO (MKE211512)
  • Alkyne-a-TCO (MKE211515)
  • Amine-a-TCO (MKE211516)

trans-Cyclooctenes (TCOs) are essential partners in the fastest known bioorthogonal reactions, but current synthetic methods are limited by poor diastereoselectivity. Especially hard to access are hydrophilic TCOs with favorable physicochemical properties for live cell or in vivo experiments. Described is a
