Myeloblastin (UniProt P24158; also known as EC 3.4.21.76, AGP7, C-ANCA antigen, Leukocyte proteinase 3, Neutrophil proteinase 4, NP-4, P29, PR-3, PR3, Wegener autoantigen) is encoded by the PRTN3 (also known as CANCA, MBN, NP4) gene (Gene ID: 5657) in human. Proteinase 3 (PR3) is a serine protease that is synthesized with a signal peptide (aa 1-25) and a propeptide (aa 26-27) that are cleaved off to produce the mature protein. It is reported to modulate the activity of platelets, endothelial cells, and inflammatory mediators, such as IL-1, IL-8, and TNF . It is a weak cationic protease that degrades elastin, fibronectin, laminin, vitronectin, and collagens I, III, and IV. Its expression is observed only in human and non-human primates and is found in azurophile granules of polymorphonuclear leukocytes and monocytes. Following its synthesis, some of it translocates to the surface of neutrophil plasma membrane in either pro- or mature form. The degree of such surface expression is genetically determined, but its surface exposure and pericellular activity can be further upregulated upon neutrophil priming and activation. Proteinase 3 autoantibody (anti-neutrophil cytoplasmic antibodies or ANCA) is the main pathogenic feature in patients suffering from granulomatosis with polyangiitis (GPA). ANCAs are shown to activate cytokine-primed neutrophils in vitro by cross-linking surface-exposed proteinase 3 and Fc receptors. Proteinase 3 activity and/or its inactivation by 1-antitrypsin varies in the human population and contributes to the risk for GPA manifestations either at onset, during relapses, or during systemic progression. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
