Quality Control Testing
Evaluated by Western Blotting in HeLa cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected TUBB6 in HeLa cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected TUBB6 in MDA-MB-231, U251-MG, NIH3T3, and PC12 cell lysates.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected TUBB6 in Human tonsil and Human fallopian tube tissue sections.
Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected TUBB6 in A431, HeLa, HUVEC, and NIH 3T3 cells.
Affinity Binding Assay: A representative lot of this antibody bound TUBB6 peptide with a KD of 3.9 x 10-7 in an affinity binding assay.

Tubulin beta-6 chain (UniProt: Q9BUF5; also known as TUBB6, Tubulin beta class V) is encoded by the TUBB6 gene (Gene ID: 84617) in human. Tubulin is the major constituent of microtubules (MTs). MTs are composed of α/β-tubulin heterodimers that self-assemble head-to-tail to form protofilaments and laterally to form a hollow tube. α/β heterodimers associate head-to-tail to form protofilaments running lengthwise along the microtubule wall with the β-tubulin subunit facing the microtubule plus end conferring a structural polarity. Tubulin binds two moles of GTP, one at an exchangeable site on the β-chain and one at a non-exchangeable site on the α-chain. Tubulin dimers constantly polymerize and depolymerize, and MTs can undergo rapid cycles of assembly and disassembly. β-Tubulin is reported to undergo phosphorylation on serine 172 by CDK1 during the cell cycle, from metaphase to telophase, but not in interphase. This phosphorylation inhibits tubulin incorporation into microtubules. TUBB6 is ubiquitously distributed with maximal expression observed in breast and lung, where it represents around 10% of all β-tubulins. High expression of TUBB6 is also observed in osteoclasts and is important for bone resorption. Its expression is largely decreased in most cancerous tissues. In malignant transformation TUBB6 has been associated with cancer invasion and metastasis in several types of aggressive malignancies. Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Avoid repeated freeze-thaws.
