Specificity

Clone 1D19 is a ZooMAb® rabbit recombinant monoclonal antibody that detects NFAT1. It targets an epitope within 19 amino acids from the N-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 19 amino acids from the N-terminal region of human NFAT1.

Application

Quality Control Testing

Evaluated by Western Blotting in Raji cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected NFAT1 in Raji cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected NFAT1 in Jurkat, and Raw264.7 lysates and 1:10,000 dilution in EL4 cell lysate.

Affinity Binding Assay: A representative lot of this antibody bound NFAT1 peptide with a KD of 6.7 x 10-9 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected NFAT1 in Jurkat cells.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected NFAT1 in human tonsil tissue sections.

Target description

Nuclear factor of activated T-cells, cytoplasmic 2 (UniProt: Q13469; also known as NF-ATc2, NFATc2, NFAT pre-existing subunit, NF-ATp, T-cell transcription factor NFAT1) is encoded by the NFATC2 (also known as NFAT1, NFATP) gene (Gene ID: 4773) in human. NFAT1 is a member of the NFAT family of transcription factors that are composed of four calcium-responsive members: NFAT1, 2, 3, and 4. They are activated by calcineurin-mediated dephosphorylation in response to increased intracellular calcium. NFAT1 is expressed in thymus, spleen, heart, testis, brain, placenta, muscle, and pancreas. Five isoforms of NFAT1 have been described that are produced by alternative splicing. Isoform 1 is highly expressed in the small intestine, heart, testis, prostate, thymus, placenta, and thyroid. Isoform 3 is highly expressed in stomach, uterus, placenta, trachea, and thyroid. NFAT1 is reported to play a role in the inducible expression of cytokine genes in T-cells, especially in the induction of the IL-2, IL-3, IL-4, TNF- . It is shown to promote invasive migration through the activation of glypican 6 expression and Wnt5A signaling pathway. In resting cells, NFAT1 is phosphorylated by NFATC-kinase on at least 18 sites in amino acids 99-363. Upon cell stimulation, all these sites except Ser-243 are dephosphorylated by calcineurin, which leads to a conformational change that simultaneously exposes an NLS and masks an NES, which results in its nuclear localization. This ZooMAbZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
