原則
葡萄糖摄取检测试剂盒(比色法)可简单、直接地测定多种细胞的葡萄糖摄取能力。该试剂盒使用葡萄糖类似物2-脱氧葡萄糖(2-DG)测量葡萄糖摄取,2-DG在测定中被细胞摄取并被己糖激酶磷酸化为2-DG6P。2-DG6P无法进入后续代谢步骤,因而会在细胞中聚集,直接成比例地反映细胞的葡萄糖摄取水平。在该测定中,2-DG的摄取水平通过偶联酶促反应方法测定,其中2-DG6P在此过程中被氧化生成NADPH,随后通过再循环扩增反应来测定NAPDH浓度,而后一反应中谷胱甘肽还原酶将利用NADPH生成谷胱甘肽。谷胱甘肽产物与DTNB反应生成TNB,后者可在412 nm波长处检测到。
