Accutase®溶液(在不含Ca ++或Mg ++的杜氏磷酸缓冲盐溶液(DPBS))包含用于细胞解离的蛋白水解酶和胶原蛋白水解酶。可用于从标准组织培养塑料器具和粘附涂层塑料器具中常规解离细胞。Accutase 是优于胰蛋白酶的细胞脱壁试剂,因为它有助于细胞从培养容器分离,损伤较少。因此,Accutase 常用于处理敏感的细胞或用于分析表面标志物。

應用

Accutase®溶液已用于将聚集簇中生长的非贴壁细胞(如脑肿瘤细胞、诱导性多能干细胞(IPSC)和胶质母细胞瘤细胞)解离或酶解成单个细胞。

适合制备测序用单细胞悬液。
证明可有效对原代成纤维细胞、内皮细胞、神经元、肿瘤细胞系和昆虫细胞进行解离。 在用于分析细胞表面标志物、病毒生长检测、流式细胞术以及生物反应器放大的细胞解离中表现尤为突出。不含有哺乳动物或细菌来源产品。

特點和優勢

公式變體

制备采用含0.5 mM EDTA•4Na以及3 mg/L酚红的Dulbecco′s PBS (0.2 g/L KCl, 0.2 g/L KH2PO4, 8 g/L NaCl以及1.15 g/L Na2HPO4)。 可参考网站上的产品数据清单获取产品用途信息。

準備報告

本品含酚红。 该产品通过干冰进行运输,包装中可能累积大量CO2。这些CO2可能会进入到溶液并略微降低pH,使溶液呈黄橘色(约pH 6.8)。这时溶液依然可正常使用,也可加氢氧化钠调节pH。一瓶完全冻住的Accutase呈黄色,解冻后变为橙色。产品打开后可重新冷冻,也可直接放于冰箱供后续使用。 不要在温水浴中解冻本品。Accutase对37 °C以上温度敏感,在37 °C下45分钟后会失活。建议放在冰箱中过夜解冻,或放在一盆冷的自来水中解冻1-1.5小时。 Accutase解冻后,摇动瓶子使溶液混匀。本品无需在使用前加热。从冰箱中取出后应立即使用,用完后放回冰箱。
