Clone 1C22 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Neprilysin/CD10. It targets an epitope within 23 amino acids from the extracellular domain.

KLH-conjugated linear peptide corresponding to 23 amino acids from the extracellular domain of human Neprilysin/CD10.

Application

Quality Control Testing

Evaluated by Western Blotting in Mouse kidney tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Neprilysin/CD10 in Mouse kidney tissue lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Neprilysin/CD10 in lysates from U87-MG cells and Human placenta and Rat kidney tissue.

Affinity Binding Assay: A representative lot of this antibody bound Neprilysin/CD10 with a KD of 4.6 x 10-8 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Neprilysin/CD10 in U-87MG cells.

Flow Cytometry Analysis: 1 µg from a representative lot detected Neprilysin/CD10 in one million Ramos cells.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected Neprilysin/CD10 in Human kidney tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Neprilysin (UniProt: P08473; also known as EC:3.4.24.11, Atriopeptidase, Common acute lymphocytic leukemia antigen, CALLA, Enkephalinase, Neutral endopeptidase 24.11, NEP, Neutral endopeptidase, Skin fibroblast elastase, SFE, CD10) is encoded by the MME (also known as EPN) gene (Gene ID: 4311) in human. Neprilysin is a single-pass type II membrane glycoprotein of the peptidase M13 family that has a cytoplasmic domain (aa 2-28), a transmembrane domain (aa 29-51), and a long extracellular domain (aa 52-750). It is highly expressed on epithelial cells of lung, intestine, kidney, breast, placenta. It displays thermolysin-like specificity but is almost confined to acting on polypeptides of up to 30 amino acids. It plays an important role in the destruction of opioid peptides such as Met- and Leu-enkephalins by cleavage of a Gly-Phe bond. It is also reported to degrade atrial natriuretic factor as well as brain natriuretic factor. Neprilysin is reported to protect neurons against amyloid peptide toxicity. It is considered as a cell surface marker in the diagnosis of acute lymphocytic leukemia. Glycosylation of Neprilysin at Asn-628 is shown to be essential for its surface expression and neutral endopeptidase activity. Mutations in MME gene have been linked to Charcot-Marie-Tooth disease that is characterized by progressive weakness and atrophy of peroneal muscles and distal muscles of the arms. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonal. (Ref.: Higuchi, Y., et al. (2016). Ann. Neurol. 79(4); 659-672; Wisner, A., et al. (2006). Proc. Natl. Acad. Sci. USA. 103(47); 17979-17984).

Forme physique

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Stockage et stabilité

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
