Elle permet de réaliser une lyse douce pour isoler les biomolécules facilement dégradables et l′ARN de bactéries. Elle a servi à la formation de sphéroplastes pour isoler l′ADN.
Purified Mutanolysin SAE0092 undergoes strict quality control testing to ensure the absence of detectable levels of contaminating DNA using 35 cycles PCR amplification of 16S and 18S rDNA using universal primer sets.
Mutanolysin is a muralytic enzyme, suitable for usage in isolation of nucleic acids, from Gram-positive bacteria (e.g. Listeria, Lactobacillus, Lactococcus),(8) and also from other bacteria that are difficult to lyse with lysozyme only.

Actions biochimiques/physiologiques

La mutanolysine est une N-acétylmuramidase. Comme le lysozyme, il s′agit d′une enzyme muralytique qui clive la liaison de la β-N-acétylmuramyl-(1→4)-N-acétylglucosamine du polymère peptidoglycane-polysaccharide des parois cellulaires bactériennes. Ses extrémités carboxyliques jouent un rôle dans la reconnaissance et la liaison de polymères uniques des parois cellulaires. La mutanolysine lyse les bactéries Listeria et autres bactéries à Gram positif comme Lactobacillus et Lactococcus.

Définition de l'unité

One unit will produce a ΔA600 of 0.01 per minute at pH 6.0 at 37 °C in a 1 mL volume using a suspension of Streptococcus faecalis cell wall as substrate.
