Description générale

Dihydropyridine Receptor (DHPR) is a heteropentamer calcium ion channel and comprises of α1,α2/δ,β and γ subunits. It is localized in the skeletal muscle in the t-tubule membrane. The α2subunit of DHPR is a glycosylated subunit and is anchored on the membrane.

Spécificité

Reacts specifically with the α2 subunit of the DHP receptor. Strong reactivity is observed with the α2 subunit in rat, mouse, guinea pig, rabbit skeletal and cardiac muscle, and rat brain and lung tissue. Weaker reactivity is observed with rat kidney, liver and spleen tissues. By immunohistochemistry, double rows of discrete punctate staining represent pairs of triads on the opposing sides of the Z-lines.

Monoclonal Anti-Dihydropyridine Receptor (α2 Subunit) antibody produced in mouse has been used in:
  • immunofluorescent histochemistry
  • surface plasmon resonance binding
  • immunohistochemistry (IHC)
  • immunoblotting

Use of this antibody for immunoblot analysis will allow for detection of a 220 kDa band (native conditions) or a 143 kDa band (denaturing conditions).Use of this antibody for immunohistochemistry of muscle tissue will demonstrate double rows of discrete punctate staining representing pairs of triads on the opposing sides of the Z-lines are observed.

Actions biochimiques/physiologiques

Dihydropyridine Receptor (DHFR) α1 subunit is the voltage-sensor. DHPR interacts with ryanodine receptor (RyR) and also mediates excitation-contraction (EC) coupling in cardiac and skeletal muscles. Mutation in the DHPR (E1014K) affects its affinity and selectivity towards calcium permeation. Additionally, it also plays a key role in the synaptic transmission.
