Musculoaponeurotic fibrosarcoma oncogene homolog B (MafB) is a transcription factor which is present in a wide variety of tissues and has 311 amino acids containing a typical bZip motif in its carboxy-terminal region.

Anwendung

The Human Protein Atlas project can be subdivided into three efforts: Human Tissue Atlas, Cancer Atlas, and Human Cell Atlas. Musculoaponeurotic fibrosarcoma oncogene homolog B (MafB) forms a homodimer through its leucine repeat structure and specifically binds to Maf-recognition elements (MAREs). It forms heterodimers with v-Maf and Fos through its zipper structure and acts as a transactivator for a promoter linked to MAREs. Through formation of numerous bZip dimers, MafB along with the AP-1 components provide great diversity in transcriptional regulation for a wide variety of genes. MafB is specifically expressed in myelomonocytic cells and binds to the DNA-binding domain of v-ets avian erythroblastosis virus E26 oncogene homolog 1 (Ets-1) by its leucine–zipper domain. It also represses Ets-1 transactivation of promoters containing Ets binding sites and inhibits Ets-1–mediated transactivation of the transferrin receptor, which is known to be essential for erythroid differentiation. Overexpression of MafB in an erythroblast cell line down-regulates the endogenous transferrin receptor gene and inhibits differentiation without affecting cell proliferation.
