General description

Estrogen receptor 1 (ESR1) encodes estrogen receptor α (ERα) protein, otherwise known as nuclear receptor subfamily 3, group A, member 1 (NR3A1), binds to estrogen. High expression of ERα is observed in tissues such as the epididymis, uterus, kidney and adrenal glands. In human chromosome, the gene ESR1 is localized on 6q25.1-q25.2.

Biochem/physiol Actions

Estrogen receptor 1 (ESR1) interacts with the nuclear transcription factor Sp1 in the presence of estradiol and regulates expression of diverse cellular and viral genes. ESR1 upon activation by estradiol interacts with transcription factor Sp3 and down regulates vascular endothelial growth factor (VEGF) in endometrial cancer cells. ESR1 tethers to DNA by protein-protein interaction with other transcription factor like activator protein 1 (AP1) family members like c-Jun, c-Fos and activating transcription factor (ATF). ESR1 and NF-κB cooperatively upregulates anti-apoptotic Baculoviral IAP (inhibitor of apoptosis protein) Repeat Containing 3 (BIRC3) and promotes breast cancer cell survival and tumor progression. Deleted in bladder cancer protein 1 (DBC1) modulates the expression of ESR1 and may promote the proliferation of breast cancer tissues. Interleukin-6 expression is suppressed by ligand activated ESR1 through interaction with NF-κB and thereby inhibits osteoporosis. ESR1 is potent in maintaining human immunodeficiency virus (HIV) latency and it is noted that latency maintenance and the amount of HIV RNA is gender specific, since women are more sensitive to estrogen.
