Culture Medium
RPMI 1640 (R0883) (with 2 mM L-Glutamine (G7513) + 25mM HEPES) + FBS / FCS (F2442), Insulin 0.6 μg/ml, Hydrocortisone 1 μg/ml, 1-Thioglycerol 10 μM.
Subculture Routine
Optimal split ratio 1:8-1:16 (seeding density 1 x 104 cells/cm2). Cells incubated at 37 °C with 5% CO2. Harvest the cells using 0.05% Trypsin/EDTA at 37 °C for 5 min. LIM2551 requires the growth medium to be changed 3 times each week. Feed and passage as for semi-adherent culture (collect medium with floating cells and then trypsinise the adherent population, pool both together before reseeding).
