Specificity

Clone 3N14 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Ataxin-2 (ATXN2). It targets an epitope within 24 amino acids from the internal region.

Immunogen

KLH-conjugated linear peptide corresponding to 24 amino acids from the internal region of human Ataxin-2 (ATXN2).

Application

Quality Control Testing

Evaluated by Western Blotting in PC12 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected ATXN-2 in PC12 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected ATXN2 in Jurkat cell lysate.

Affinity Binding Assay: A representative lot of this antibody bound ATXN2 peptide with a KD of 6.1 x 10-7 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected ATXN2 in Human cerebral cortex tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected ATXN2 in PC12 cells.

Target description

Ataxin-2 (UniProt: Q99700; also known as Spinocerebellar ataxia type 2 protein, Trinucleotide repeat-containing gene 13 protein) is encoded by the ATXN2 (also known as ATX2, SCA2, TNRC13) gene (Gene ID: 6311) in human. ATXN-2 is a highly conserved, monomeric, eukaryotic RNA-binding protein that is ubiquitously expressed, and its higher expression is observed in the mammalian nervous system. It can also form homodimers. Its expression is reported to increase with age in Purkinje cells. At the subcellular level, it is reported to localize to the nucleolus, nucleoplasm, Golgi apparatus, rough ER membranes, and the receptor endocytosis machinery at the plasma membrane. It carries an Sm-domain (aa 267-344) and a PAM-2 motif on the C-terminal side of the Sm-domain. The PAM-2 motif mediates its interaction with the C-terminal helical domain of the poly(A) binding protein. ATXN2 and its associated protein complex are reported to control distinct steps in post-transcriptional gene expression, including poly-A tailing, RNA stabilization, microRNA-dependent gene silencing, and translational activation. Mutations in ATXN2 gene cause several diseases and/or modifies several and disparate neurological disorders. Some mutations are linked to development of Spinocerebellar ataxia 2 (SCA2), a genetically heterogeneous group of cerebellar disorders that lead to poor coordination of hands, speech, and eye movement. Five isoforms of ATXN-2 have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
