Specificity

Clone 1I5-H2 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Collectin-11. It targets an epitope within 17 amino acids from the C-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 17 amino acids from the C-terminal half of human Collectin-11.

Application

Quality Control Testing

Evaluated by Western Blotting in Human liver tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Collectin-11 in Human liver tissue lysate.

Tested Applications

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Collectin-11 in Human kidney tissue sections.

Immunofluorescence Analysis: A 1:100 dilution from a representative lot detected Collectin-11 in Human kidney tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound Collectin-11 peptide with a KD of 1.0 x 10-12 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Collectin-11 (UniProt: Q9BWP8; also known as Collectin kidney protein 1, CL-K1) is encoded by the COLEC11 gene (also known as UNQ596/PRO1182) gene (Gene ID: 78989) in human. Collectin-11, a disulfide-linked, calcium-dependent, homotrimeric protein of the lectin family of pattern recognition molecules, is synthesized with a signal peptide (aa 1-25), which is subsequently cleaved off to produce the mature protein. It contains a collagen-like domain (aa 65-112) and a C-type lectin domain (aa 149-265). It is also reported to form heteromeric complexes with Collectin Liver 1 (CL-10) in the serum. Collectin-11 is most obviously expressed in renal tubules by epithelial cells. It displays antimicrobial properties and has ability to trigger complement activation via the lectin pathway. It plays a role in innate immunity through its ability to bind non-self-sugars presented by microorganisms and to activate the complement through the recruitment of Mannan-binding lectin-associated serine protease 1 (MASP-1). Its structure is similar to other C-type lectins and consists of a globular head followed by a neck and a collagenous tail. The head region contains a carbohydrate recognition domain (CRD), and the tail contains binding sites for MASPs. Collectin-11 monomers form a triplet structure that self-combines to form oligomers with higher avidity of binding to ligand. Ten isoforms of Collectin-11 have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
