Specificity

Clone 2O20 is a Rabbit recombinant monoclonal antibody that specifically detects NLR family CARD domain-containing protein 3 (NLRC3/NOD3). It targets an epitope within 15 amino acids from the N-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 15 amino acids from the N-terminal region of human NLR family CARD domain-containing protein 3 (NLRC3/NOD3).

Application

Quality Control Testing

Evaluated by Western Blotting in Jurkat cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected NLRC3/NOD3 in Jurkat cell lysate.

Tested applications

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected NLRC3/NOD3 in human spleen tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected NLRC3/NOD3 in Jurkat cells.

Affinity Binding Assay: A representative lot of this antibody bound NLRC3 with a KD of 1.3 x 10-7 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

Target description

NLR family CARD domain-containing protein 3 (UniProt: Q7RTR2; also known as CARD15-like protein, Caterpiller protein 16.2, CLR16.2, NACHT, LRR and CARD domains-containing protein 3, Nucleotide-binding oligomerization domain protein 3) is encoded by the NLRC3 (also known as NOD3) gene (Gene ID: 197358) in human. NLRC3/NOD3 is a cytoplasmic protein that serves as a negative regulator of innate immune response. It attenuates signaling pathways activated by Toll-like receptors (TLRs) and the DNA sensor STING/TMEM173 in response to pathogen-associated molecular patterns, such as intracellular poly(dA:dT), but not poly(I:C), or in response to DNA virus infection. It may affect TLR4 signaling by acting at the level of TRAF6 ubiquitination, decreasing the activating lysine 63-linked ubiquitination and leaving unchanged the degradative lysine 48-linked ubiquitination. In primary T-cells, it is down-regulated upon T-cell receptor activation. It is reported to inhibit the PI3 kinase-AKT-mTOR pathway possibly by directly interacting with the p85 regulatory subunit of PI3 kinase and disrupting the association between the regulatory (p85) and the catalytic (p110) subunits. By regulating PI3 kinase-Akt-mTOR pathway, it can exert its control on cell proliferation. NLRC3/NOD3 levels are shown to be significantly reduced in colorectal cancers. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
