General description

Anti-GABA is produced in rabbit using GABA-BSA as the immunogen. The antibody is isolated from antiserum by immunospecific methods of purification. Antigen specific affinity isolation removes essentially all rabbit serum proteins, including immunoglobulins which do not specifically bind to GABA.
GABA (γ-aminobutyric acid) is a multifunctional molecule found in several organisms, from prokaryotes to vertebrates. It is present in non-nervous structures, such as peripheral nervous and endocrine systems. GABA is formed following decarboxylation of glutamic acid by the enzyme glutamic acid decarboxylase (GAD).

γ-aminobutyric acid (GABA) conjugated to BSA.

Application

Expression of GABA in neucortical cells harvested from the brains of E19 day old rat embyros was detected by immunofluorescence using rabbit anti-GABA antibody. Triple IF staining was performed with the anti-GABA antibody and two anti-GAD antibodies. Expression of GABA was analyzed in cells isolated from the pallium of various animals including rats, mice, rabbits, guinea pigs, and lizards by immunohistochemistry. IHC was performed using rabbit anti-GABA antibody at 1:1000 diluted in a solution of 0.01M PBS ph 7.4 + 0.5% triton-x100.
Rabbit anti-GABA antibody has been used for electron microscopy analysis in rat hippocampal tissues at a dilution of 1:4000. The antibody has also been used for immunocytochemistry applications at dilutions ranging from 1:100-1:750 in Drosophila and Periplaneta americana brain cells.

Biochem/physiol Actions

GABA (γ-aminobutyric acid) is the major inhibitory neurotransmitter in the central nervous system and also functions as a neuromodulator in some peripheral tissues. It controls heart rate, blood pressure and respiratory process. GABA facilitates neuronal inhibition by interacting with GABA/benzodiazepine receptor and opening an integral chloride channel. Rabbit anti-GABA antibody shows positive binding with GABA and GABA-keyhole limpet hemocyanin (KLH) in a dot blot assay, and exhibits negative binding with bovine serum albumin (BSA).

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 1% BSA and 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2-8 °C for up to one month. For extended storage, solution may be frozen in working aliquots. Repeated freezing and thawing is not recommended. If slight turbidity occurs upon prolonged storage, clarify by centrifugation before use.
