General description

Propidium iodide (PI) is often used for cell cycle analysis by flow cytometry. Compared to this analysis method, the BioTracker Far Red Live Cell Cycle Assay Solution does not require cell fixation or permeabilization, and is permeable in live cells. Use 5 uL of probe solution in 500 uL PBS for each sample. Vial contains enough for 50 tests.The far red probe uses the 633 laser, enabling multiplexing with shorter wavelength probes by freeing the 488 laser for probes that emit in the green range of the spectrum. IMPORTANT APPLICATION NOTES:*Wash cells thoroughly to remove trypsin. Residual trypsin can confound analysis of flow cytometry populations.*Adjust gates after cells are stained with probe SPECTRAL PROPERTIESExcitation: 633 - 647 nmEmission: 695 LP or 780/60 BP.A filter set for APC-Cy7 is suitable.

Target description

The control system of the cell cycle is closely linked to cell proliferation. Cell division in which one cell produces two daughter cells occurs as part of the cell cycle. The cell cycle can be assessed by analyzing cells stained for DNA using flow cytometry, followed by analysis of the proportion of stained nuclei in cell cycle phase. The cell cycle is classified into two major phases: interphase and the mitotic (M) phase. Interphase is composed of G1 phase (preparation for DNA synthesis which occurs prior to cell division), S phase (DNA synthesis and replication), and G2 phase (a single cell with duplicated chromosomes). M phase involves mitosis and cytokinesis. Cell cycle progression is regulated by cyclins and cyclin-dependent kinases. To determine the action of anticancer drugs and others, it is important to analyze the mechanism of cell-cycle checkpoint.

Live cell probe for fluorescent cell cycle analysis. Unlike propidium iodide, this probe is membrane permeable and highly specific for DNA, which makes it possible to analyze the cell cycle by simply adding the reagent to a cell suspension.
