General description
Protein arginine N-methyltransferase 7 (EC 2.1.1.321; UniProt Q9NVM4; also known as Histone-arginine N-methyltransferase PRMT7, [Myelin basic protein]-arginine N-methyltransferase PRMT7) is encoded by the PRMT7 (also known as KIAA1933) gene (Gene ID 54496) in human. Mammalian protein arginine methyltransferases (PRMTs) catalyze the methylation of arginine residues on histone and non-histone proteins. PRMTs are divided into three types based on their mode of activity. Type I PRMTs (PRMT1–4, PRMT6, and PRMT8) transfer up to two methyl groups on the same terminal guanidino nitrogen of arginine to form ω-NG-monomethylarginine (MMA) and ω-NG,NG-asymmetric dimethylarginine (ADMA) residues. Type II PRMTs (PRMT5 and PRMT9) transfer methyl groups on different terminal guanidino nitrogens to form ΜΜΑ and ω-NG,N′G-symmetric dimethylarginine (SDMA) residues, while type III PRMT7 transfers only one single methyl group to form MMA residues. PRMT7 is involved in mediating cellular resistance or sensitivity to cytotoxic agents, as well as in regulating male germline imprinted gene via interaction with CCCTC-binding factor-like protein (CTCFL). PRMT7 is also reported to inhibit neuronal cell differentiation through the repression of MLL4-dependent gene expression, implicating a role in a stem cell population maintenance and cancer development. PRMT7 knockdown is shown to decrease breast cancer cell invasion in vitro and metastasis in vivo, while PRMT7 overexpression is shown to enhance the invasiveness of a non-aggressive breast cancer cell line by inducing the expression of MMP9. Similar to PRMT9, but unlike other PRMTs, PRMT7 contains a duplicated methyltransferase domain (a.a. 14-345 and 358-684), with the N-terminal MTase acting as a funtional AdoMet-binding domain and the C-terminal MTase domain serving a pseudodimer function, mimicking the homodimer formation of other PRMTs needed for subtrate processing. Deletion of either domain in PRMT7 abolishes its activity.
Specificity
Clone 7G1 was raised against the full-length 692-a.a. spliced isoform (UniProt Q9NVM4-1), reactivity toward the other three spliced isoforms reported by UniProt is possible, but has not been confirmed.
