General description

DAX-1 is an orphan nuclear receptor and is required for the normal development of the hypothalamic-pituitary-adrenal-gonadal axis formation. Defects in DAX-1 are the cause of X-linked adrenal hypoplasia congenital (XL-AHC); also known as X-linked Addison disease (AHX). DAX-1 also has a role in the development of the embryo and in the maintenance of embryonic stem cell pluripotency by acting as a negative regulator of Oct3/4 in ES cells.

Immunogen

Linear peptide corresponding to Human DAX-1 (amino acids 135-166).

Application

Anti-DAX-1, clone 2F4, has been shown to work in Immunocytochemical (Lehmann et al., 2009) and Immunohistochemical (Ehrlund et al., 2013) analysis.

This Anti-DAX-1, clone 2F4 Antibody is validated for use in Western Blotting and Immunocytochemistry and Immunohistochemistry for the detection of DAX-1.

Quality

Evaluated by Western Blot in a Dax-1 overexpressing HEK293 transfected cell line. Western Blot Analysis: A 1:1000 dilution of this antibody detected DAX-1 in a DAX-1 overexpressing HEK293 transfected cell line.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.

In recent studies, we have found that DAX-1 (dosage-sensitive sex reversal/adrenal hypoplasia congenita critical region on the X chromosome) is expressed in the mouse mammary epithelial cell line HC11. In this study, we focused on the regulation of DAX-1 expression

Mutations in the human DAX-1 gene lead to X-linked adrenal hypoplasia congenita and hypogonadotropic hypogonadism. DAX-1 has been proposed to play a role in steroidogenesis because it is highly expressed in adrenocortical and testicular Leydig cells and because loss-of-function mutations

Genetic and experimental evidence points to a critical involvement of the atypical mammalian orphan receptor DAX-1 in reproductive development and steroidogenesis. Unlike conventional nuclear receptors, DAX-1 appears not to function as a DNA-bound transcription factor. Instead, it has acquired the
