General description

CB1 is a protein that is one of two cannabinoid receptors (CB1 CB2). Cannabinoid principally delta-9-tetrahydrocannabinol (THC) and its synthetic analogs, are the active psychoactive compounds present is marijuana. These cannabinoid receptors are members of the guanine-nucleotide-binding protein (G-protein) coupled receptors which inhibit adenylate cyclase activity in a dose-dependent, stereoselective and pertussis toxin-sensitive manner. The two receptors have been found to be involved in the cannabinoid-induced CNS effects (including alterations in mood and cognition) experienced by users of marijuana. Three splice variants have been found for this gene. CB1 is primarily located in brain, and CB2 is found macrophages.

Specificity

Recognizes rat Cannabinoid Receptor 1 (CB1). The immunogen shows no significant sequence homology with CB2 or other receptors.

SPECIES REACTIVITIES: The immunogen peptide is 100% conserved mouse, 94% in monkey and human, 88% in zebra finch, 75% in feline and newt, (salamander) and 73% in puffer fish. Reactivity with other species has not been confirmed.

A 20 amino acid peptide sequence near the cytoplasmic, C-terminus of rat CB1 (Gerard et al. 1991; Gerard et al.

1990; Shire et al. 1995; Matsuda et al. 1990; Ho et al. 1996; Chakraborty et al. 1995).

Western blot: 1-10 μg/mL using ECL. Expected sizes are ~60kDa and 80-84kDa under reducing conditions.

Immunohistochemistry: not tested. It is recommended that the antibody be tried at 2-10 μg/mL on paraformaldehyde fixed tissue.

ELISA: 1:10,000-1:100,000 using 50-100 ng control peptide (Catalog number AG213) per well.

Optimal working dilutions must be determined by the end user.

Physical form

Affinity purified immunoglobulin. Liquid in phosphate buffered saline containing 0.1% BSA.

Storage and Stability

Maintain at -20°C in undiluted aliquots for up to 6 months. Avoid repeated freeze/thaw cycles.
