General description

Mouse Connexin 40 is a 358 amino acid gap junction protein with a predicted M.W. of ~40 kDa. It is prominently expressed in lung, heart and skin (see reviews: Kumar & Giula 1996; White, 1995; Evans, 1994; Beyer , 1990; Henneman, 1992; Traub, 1994; Goliger & Paul 1994). One gap junction consists of a cluster of closely packed pairs of transmembrane channels, the connexons, through which materials of low MW diffuse from one cell to a neighboring cell. A connexon is composed of a hexamer of connexins.

Specificity

Reactivity with other species has not been determined.
Reacts with mouse connexin 40.

Immunogen

Anti-Connexin 40 is made against a 19 amino acid peptide sequence within the C-terminal cytoplasmic domain of mouse C×40 (2). Peptide sequence is conserved in human and rat Cx40 (100%), chicken Cx42 (77%), human Cx50 (87%), rat Cx50 (81%) and sheep Cx49 (87%).

Application

Anti-Connexin 40 Antibody is an antibody against Cx40 for use in IHC & WB.
Immunohistochemistry Analysis: An 1:50 dilution of a representative lot detected Connexin 40 immunoreactivity associated with the gap junction between the cardiac myoepithial cells in mouse heart tissue sections.
Western Blot Analysis: An 1:1000 dilution of a representative lot detected Connexin 40 in human (HEK293, HeLa, HepG2, HUVEC, Jurkat) and murine (C2C12, HIN/3T3) cell lysates.
Western Blot Analysis: An 1:000 dilution of a representative lot detected Connexin 40 in human, mouse, and rat heart tissue homogenates, as well as in mouse brain tissue homogenates.

Optimal working dilutions must be determined by end user.

Quality

Evaluated by Western Blot on Jurkat lysates.

Western Blot Analysis: A 1:1000 dilution of this lot detected Connexin 40 in 10 μg of Jurkat lysates.

Gap junctions are involved in vascular growth and their expression pattern is modulated in response to hemodynamic conditions. They are clusters of intercellular channels formed by connexins (Cx) of which four subtypes are expressed in the cardiovascular system, namely Cx37

The endothelium-derived hyperpolarizing factor (EDHF) contributes critically to the regulation of vascular tone. Its dependency on direct signaling through myoendothelial gap junctions composed of connexins (Cx) is controversially discussed. We studied the impact of Cx40 in EDHF-type dilations in vivo
