生化/生理作用
环孢素A(cyclosporin A)通常在移植术后给药,用于抗排斥反应。在对器官移植起效之外,环孢素A(CyA)也会带来肾毒性副作用。
环孢素A(cyclosporin A, CsA)是由多孔木霉(Tolypocladium inflatum)真菌合成的非极性环状多肽。它是一种强效的免疫抑制剂,主要作用于T淋巴细胞,已证明可在mRNA转录水平抑制多种核蛋白的T细胞活化作用。 它可与胞内受体亲环素形成复合物,复合物可结合一种钙离子/钙调依赖性蛋白磷酸酶——钙调神经磷酸酶(calcineurin),抑制其酶活性。 CsA可抑制培养肝细胞中丙型肝炎病毒基因组的复制。 > 10 nM浓度的CsA可保护肝细胞免遭鬼笔环肽破坏。 CsA可通过抑制钙调神经磷酸酶抑制T细胞活化生产 IL2 。
