胰岛素-转铁蛋白-硒-乙醇胺添加剂(100×SITE液体培养基补充剂)是适宜的无血清培养基替代品,含有适合体外生长和分化研究的纯净因子。补充剂在培养基中的添加量并不一定,取决于研究细胞类型和采用的基础培养基。它是一种通用的细胞补充剂,设计用于含有丙酮酸钠的非复合型培养基(如最低必需培养基(MEM)、Roswell Park Memorial Institute培养基(RPMI-1640))及复合型培养基(如Ham′s F-12、杜氏改良Eagle培养基(DME)/F-12、MEM)。

應用

胰岛素-转铁蛋白-硒-乙醇胺添加剂(100×SITE液体培养基补充剂)已用于:
  • 小鼠胚胎胰腺上皮细胞3D培养
  • 兔胃腺和胃壁细胞分离
  • 兔肾近端小管上皮细胞分离

生化/生理作用

  • 胰岛素是一种多肽激素,可促进葡萄糖和氨基酸的摄取,这种特性可能与观察到的有丝分裂作用有关。
  • 转铁蛋白:铁运输蛋白。铁是一种必需微量元素,但游离形式可能有毒。为了保证细胞培养的营养,可与血清转铁蛋白结合后使用。
  • 硒:通常由血清提供的必需微量元素。
  • 乙醇胺:对杂交瘤细胞增殖起重要作用,这类细胞的培养基补充剂中常添加乙醇胺。

成分

含有1.0mg/ml人重组胰岛素,0.55mg/ml 人转铁蛋白(基本上不含贴),0.5μg/ml 亚硒酸钠,以及 0.2mg/ml 乙醇胺,100x 浓缩液。
