磷酸甘油酸激酶(PGK)是一种糖酵解酶,可从多种生物体中分离获得。这种典型的铰链弯曲单体酶在三域生物中都很保守。这种酶由单体折叠多肽链构成,可分成两个几乎相同的结构域,每个结构域由两个螺旋链(螺旋链7和14)连接并由一个深裂隙分离。这种排列使该酶具有独特的双叶结构。

應用

来自面包酵母的′3-磷酸甘油酸磷酸激酶(酿酒酵母)已用于:

生化/生理作用

3-磷酸甘油酸磷酸激酶催化磷酸基团从1,3-二磷酸甘油酸到ADP的可逆转移,以生成ATP和3-磷酸甘油酸。 3-磷酸甘油酸磷酸激酶的活性对糖酵解和糖异生作用是必需的。磷酸甘油酸激酶(PGK)通过催化两个产生ATP反应之一的糖酵解途径中发挥着重要的作用。它将1,3-二磷酸甘油酸(1,3BPGA)转化为3-磷酸甘油酸(3PGA)。此外,它还通过催化对峙反应生成1,3BPGA和二磷酸腺苷(ADP)参与糖异生过程。由于其参与除能量代谢之外的许多过程,如发病机制、与核酸的相互作用、肿瘤发生和发展、细胞死亡及病毒复制等,PGK还被称为兼职蛋白质。
