胍硫氰酸酯是一种离液剂，可用作强蛋白质变性剂。它被广泛应用于 RNA 分离方案。胍硫氰酸酯能不可逆地灭活 RNase 和 DNase 并破坏细胞膜。它通过扰乱蛋白质的二级结构来溶解蛋白质。胍硫氰酸酯还能将核蛋白从核酸中分离出来，并有助于蛋白酶对蛋白质的消化。它提高了蛋白酶 K 的活性，使其成为首选蛋白酶。

應用

胍硫氰酸酯溶液已用于提取：
  • 人类免疫缺陷病毒（HIV）感染和对照患者的血浆 RNA
  • 从收集的组织（芽、花、果实和种子）中提取总 RNA
  • 在逆转录聚合酶链反应（RT-PCR）分析中，从畸胎瘤细胞系（Tera-1）提取 RNA

注意

稍微加热至40-50℃，可能结晶的产物会再次溶解。
