Cell Line Origin

Mouse BALB/c (H-2d) T cells x Mouse AKR Thyoma, BW5147

Cell Line Description

Do-11-11 was produced by fusion of the antigen-specific I region-restricted (H-2d) BALB/c T-cell blasts with the AKR thyoma BW5147. This murine helper T cell hybridoma is specific for chicken ovalbumin (cOva) restricted to I-Ad and cross reacts with cOva I-antibody. The hybrid can be stimulated to secrete Interleukin 2 (IL-2) or to act as an antigen-specific helper T-cell. Expression of IL-2 can be lost by long-term culture of growth at high densities.

Culture Medium

DMEM or RPMI 1640 + 2 mM Glutamine + 10% Fetal Bovine Serum (FBS).

Subculture Routine

Maintain cultures between 3-9 x 100,000 cells/ml; 5% CO2; 37 °C. Cells should not be grown at high density; avoid long-term culture as this might lead to change of cell receptors.
