Specificity

Clone 1B5 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Cyclic GMP-AMP synthase (cGAS). It targets an epitope within 22 amino acids from the N-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 22 amino acids from the N-terminal region of human Cyclic GMP-AMP synthase (cGAS).

Application

Quality Control Testing

Evaluated by Western Blotting in Raw264.7 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected cGAS (Cyclic GMP-AMP synthase) in Raw264.7 cell lysate.

Tested applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected cGAS in A549 and MCF-7 cell lysates.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected cGAS in human placenta tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound cGAS peptide with a KD of 1.0 x 10-12 in an Affinity Binding Assay.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected cGAS in MCF-7 cell line.

Flow Cytometry Analysis: 1 μg from a representative lot detected cGAS in one million A549 cells.

Anti-cGAS, clone 1B5 ZooMAb®, Cat. No. ZRB1406, is a recombinant Rabbit monoclonal antibody that targets cGAS and is tested for use in Affinity Binding Assay, Flow Cytometry, Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.

Target description

Cyclic GMP-AMP synthase (UniProt: Q8N884; also known as EC:2.7.7.86, cGAMP synthase, cGAS, h-cGAS, 2′3′-cGAMP synthase, Mab-21 domain-containing protein 1) is encoded by the CGAS (also known as C6orf150, MB21D1) gene (Gene ID: 115004) in human. cGAS is a nucleotidyltransferase that catalyzes the formation of cyclic GMP-AMP (cGAMP) from ATP and GTP and plays a key role in innate immunity. In resting conditions, it is localized at the cell membrane as a peripheral membrane protein by binding to phosphatidylinositol 4,5-bisphosphate (PtdIns(4,5)P2). Membrane localization is reported to be essential to limit the recognition of self-DNA. Upon detection of double-stranded DNA (dsDNA), it is released from the cell membrane into the cytosol to commence a signaling process. Its phosphorylation at tyrosine 215 by B-lymphocyte kinase (BLK) is reported to promote its cytosolic retention. Following dephosphorylation it translocates to the nucleus. cGAS can undergo acetylation at lysine 384, 394, and 414, which inhibits its activity. It acts as a key cytosolic DNA sensor. It binds to cytosolic DNA directly, leading to activation and synthesis of cGAMP, a second messenger that binds to and activates TMEM173/STING, thereby triggering type-I interferon production. It can also act as an innate immune sensor of infection by retroviruses, such as HIV-1, by detecting the presence of reverse-transcribed DNA in the cytosol and can also detect the presence of DNA from bacteria, such as Mycobaterium tuberculosis. In addition to its antiviral activity, it is also involved in the response to cellular stresses, such as senescence, DNA damage or genome instability. It acts as a suppressor of DNA repair in response to DNA damage and translocates to the nucleus following dephosphorylation at tyrosine 215 and inhibit homologous recombination repair by interacting with PARP1. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
