Application

All Prestige Antibodies Powered by Atlas Antibodies are developed and validated by the Human Protein Atlas (HPA) project and as a result, are supported by the most extensive characterization in the industry.

The Human Protein Atlas project can be subdivided into three efforts: Human Tissue Atlas, Cancer Atlas, and Human Cell Atlas. The antibodies that have been generated in support of the Tissue and Cancer Atlas projects have been tested by immunohistochemistry against hundreds of normal and disease tissues and through the recent efforts of the Human Cell Atlas project, many have been characterized by immunofluorescence to map the human proteome not only at the tissue level but now at the subcellular level. These images and the collection of this vast data set can be viewed on the Human Protein Atlas (HPA) site by clicking on the Image Gallery link. Biochem/physiol Actions

Caspase 8 (CASP8) is the first protease of the activation cascade of caspases responsible for the tumor necrosis factor receptor superfamily member 6 (TNFRSF6/FAS) mediated and tumor necrosis factor receptor superfamily member F1A (TNFRSF1A) induced cell death. After binding to the adapter molecule, Fas-associated via death domain (FADD) recruits it to a receptor. This results in the formation of an aggregation called death-inducing signaling complex (DISC) which carries out the proteolytic activation of CASP8. Then its active dimeric form is liberated from the DISC and activates downstream apoptotic proteases. It then cleaves and activates CASP3, CASP4, CASP6, CASP7, CASP9 and CASP10. CASP8 also participates in Granzyme (GZMB) apoptotic pathways. It hydrolyzes a small-molecule substrate, Ac-Asp-Glu-Val-Asp-AMC.CASP8 is a modulator of p85a Rab-GAP activity and endosomal trafficking. The interaction between it and p85a causes Rab5 GTP loading, changes endosomal trafficking and at the edge of the cell, Rab5-positive endosomes are seen.
