應用

胰蛋白示磷酸盐培养基可用作以下物质的组成成分:
  • 用于培养BME26蜱胚细胞系的Leibovitz L-15培养基
  • 用于制备鸡胚纤维母细胞(CEF)的M199培养基
  • 用于转染前培养幼仓鼠肾(BHK-21)的Glasgow′极限必需培养基(GMEM)

除了用于苛养菌的生长外,还研究了胰蛋白胨磷酸盐肉汤(TPB)作为补充剂,用于制备能够支持在高密度灌注培养搅拌槽生物反应器中BHK-21细胞的疫苗生产以及SF21昆虫细胞生长的培养基。

生化/生理作用

胰蛋白胨磷酸盐肉汤含有氨基酸类营养素和存活因子,能够支持布鲁氏菌(Brucella)、链球菌(Streptococcus)和奈瑟氏球菌(Neisseria)等苛养菌的生长,以及昆虫和动物细胞等真核细胞的生长。葡萄糖提供可发酵的碳水化合物,以供苛养菌使用。氯化钠保持渗透和离子平衡,磷酸二钠提供基本的缓冲能力。

成分

胰蛋白胨磷酸盐肉汤(TPB)由四种成分组成:胰蛋白胨(20g/L),葡萄糖(2g/L),NaCl(5g/L)以及磷酸二钠(2.5g/L),通常调节至pH 7.3。胰蛋白胨组分是一种蛋白胨(氨基酸和短肽的混合物),来源于乳蛋白酪蛋白的混合酶水解物(胰酶)。该水解物含有氨基酸类营养素和存活因子,能够支持布鲁氏菌、链球菌和奈瑟氏球菌等苛养菌的生长,以及昆虫和动物细胞等真核细胞的生长。葡萄糖提供可发酵的碳水化合物,以供苛养菌使用。氯化钠保持渗透和离子平衡,磷酸二钠提供基本的缓冲能力。
