IgG抗体亚型是免疫系统中含量最高的血清免疫球蛋白。它由B细胞分泌,存在于血液和细胞外液中,可防止细菌、真菌和病毒引起的感染。母体IgG可通过胎盘转移至胎儿,胎盘对新生儿免疫防御感染至关重要。
小鼠IgG(全分子)-TRITC抗体是小鼠IgG和所有亚类的特异性抗体,会与小鼠IgA和IgM交叉反应。抗体采用离子交换色谱法进行纯化,然后与四甲基罗丹明异硫氰酸酯(TRITC)结晶偶联。

对完整的抗血清进行分级,然后采用离子交换色谱法进一步纯化,得到抗血清的IgG级分。该部分基本上不含其他山羊血清蛋白。将该抗体与异硫氰酸四甲基罗丹明(TRITC)结晶缀合,然后进一步纯化除去游离的TRITC。 采用Ouchterlony双向扩散(ODD)法测定小鼠IgG的特异性。抗体制备对小鼠 IgG 和所有亚类均具有特异性,其与小鼠 IgA 和 IgM 发生交叉反应。偶联前,通过免疫电泳(IEP)鉴定抗体及其纯度。 对该抗体制剂进行电泳,然后与山羊IgG抗体和抗山羊全血清进行扩散,会在γ区形成沉淀弧。

應用
小鼠IgG(全分子)-TRITC抗体可用于小鼠脾细胞的免疫荧光,最低抗体稀释度为1:64。在各种研究中,免疫荧光法使用的抗体稀释范围为1:50至1:250。
