Analyze Iron (Total) in Plant Material

The Spectroquant® Iron test allows the accurate quantification of iron ions in various waters. All iron ions are reduced to iron(II) ions by ascorbic acid. In a thioglycolate-buffered medium, these react with a triazine derivative to form a red-violet complex that is measured photometrically.

Materials

Plant Material Sample Preparation

Ground the plant material finely and pass through a sieve (mesh 0.4 - 0.7 mm) and dry the material at 105 °C for 3 hours.

Add 1.0 g of the dried material to a mixture of 1.5 mL nitric acid 65 %, 0.5 mL perchloric acid 70 %, 1 mL sulfuric acid 95 - 97 % and heat until white fumes are envolved (fume cupboard!). Dissolve the residue, after cooling, in water for analysis and fill up to the mark in a 100-mL standard volumetric flask and mix well.

Analysis

Determine iron content with the above mentioned test kits.
